I'm running Avast Anti-Virus on one comp and Microsoft Security Essentials on the other.

Try 0.5.3 (just out).
Remove the INI file and walk through the Wizard.
Other than that, I'm out of suggestions.

I had this same problem and figured it out.

I had a crack change my hosts file entry for 'localhost' to 0.0.0.0 instead of 127.0.0.1... changing it back fixed it
