I installed SABnzbd on Proxmox on a server. It downloads to a mounted drive to a NAS (temporary/complete folder).
To save energy I am trying to shut the Synology down during the night and when no one is using it.
But whenever the Synology is shut down, SABnzbd loses connection if already opened in the browser (Lost connection to SABnzbd.. In case of restart this screen will disappear automatically!) or cannot be accessed at all.
As soon as the Synology is back online everything works again.
Does SAB check, whether the download folder is available and if not, produce this error message?
The logs don't show any error message.
DEBUG only logs the attempted API connection from my browser.
Is there any other setting which could produce this error?

Check your connection in Status and Interface settings window.
Use Test Server in Config > Servers.
We will probably ask you to do a test using only basic settings. - Do you experience problems during repair or unpacking?
Enable +Debug logging in the Status and Interface settings window and share the relevant parts of the log here using [ code ] sections.

Yes, there is a diskspace check thats performed every so often.
We check during startup if your disks are present and can even wait for them to come online.
However, we don't support the Temporary directory not being present since we rely on it being there.
I would suggest you instead run SABnzbd on the NAS itself, we provide a Synology package for that purpose.

So it should work if I were to change the temporary directory to the local storage instead of the NAS and move Files after extraction?

I would try to leave Temp on your local drive but then put Complete on the NAS. Then just use the SABnzbd scheduler to pause while your NAS is turned off.
I think your interface should stay available and they still unpack to NAS.
